5.Puncture the follicles with a 27 gauge syringe needle in order to release the external GCs and cumulus-oocyte complexes (COCs), usually loosely packed into the follicles. Use COCs for IVM.

6.Bisect the remnant of follicles immersed in a petri dish containing 2 ml BSA-CM supplemented with 50 UI/ml heparin.

7.Remove cells attached to the follicle wall by scraping with a fine plastic loop (primarily mural GCs) under a stereomicroscope.

8.Recover collection media containing GCs in a 2 ml Eppendorf microtube and spin at 1200 × g for 2 min at room temperature.
